It represents the last meal shared by Jesus with his 12 disciples before his capture and the Crucifixion. It shows specifically the moment when Jesus has just said "one of you will betray me".
The leftmost threesome staring to the right together with the rightmost pointing to the left gravitate to the central. The second triplet on the right is shocked and angry, leaning forward. The second triplet on the left is feeling suspicious or guilty, leaning away. Jesus is detached and calm, whilst Judas is holding on to his moneybag.
The mural is in the refectory of the Convent of Santa Maria della Grazie ("Holy Mary of Grace") in Milan:
The finished wall painting deteriorated rapidly, because da Vinci had used tempera over gesso ground, instead of using the reliable technique of fresco.
When King Louis XII of France fell Milan and saw this fresco, he desired to bring it back to France, but in vain.

According to the New Testament, John the Baptist was Jesus Christ's cousin. Anne was mother of Mary.
